Resumen Introducción: El embalse Cerrón Grande de 135 km2 es el humedal más grande de El Salvador, reconocido como "Humedal de Importancia Internacional" por la Convención Ramsar. Además, es uno de los ecosistemas más contaminados a nivel nacional, enfrentando serios problemas, entre ellos, las proliferaciones de cianobacterias. Objetivo: Evaluar las relaciones entre los factores ambientales y la distribución espacial y temporal de las cianobacterias en el embalse a lo largo de un año hidrológico. Métodos: Extrajimos agua con cianobacterias de seis sitios fijos, desde octubre 2018 hasta septiembre 2019. Resultados: Identificamos 16 géneros, el más abundante Microcystis sp. Además, Dolichospermum sp.; Microcistis sp. alcanzaron 1.5 x 106 células/ml en junio 2019, principalmente en el sector noroeste, que es poco profundo y cercano a las zonas de confluencia de afluentes altamente contaminados que llegan al embalse. El sector sureste más profundo tenía bajas concentraciones de estos organismos. Los factores ambientales mayormente relacionados con dicho comportamiento fueron: fósforo, la conductividad eléctrica y el valor de Secchi, como se esperaba de un cuerpo eutrófico. Conclusiones: La variación en la concentración celular de cianobacterias en este embalse está altamente influenciada por la lluvia, la mezcla de agua y el contenido de nutrientes.


Abstract Introduction: The 135 km2 Cerrón Grande reservoir is the largest wetland in El Salvador, recognized as "Wetland of International Importance" by the Ramsar Convention. It is also one of the most polluted ecosystems nationwide, facing serious problems, among them, a proliferation of cyanobacteria. Objective: To assess the relationships among environmental factors and the spatial and temporary distribution of cyanobacteria in the reservoir throughout a hydrological year. Methods: We extracted water with cyanobacteria from six fixed sites, from October 2018 through September 2019. Results: We identified 16 genera, the most abundant Microcystis sp. And Dolichospermum sp.; Microcystis sp. reached 1.5 x 106 cells/ml in June 2019, mainly in the north-west sector, which is shallow and close to the confluence zones where highly polluted tributaries reach the reservoir. The deeper south-east sector had low concentrations of these organisms. The environmental factors mostly related to this behavior were: phosphorus, electrical conductivity and Secchi value, as expected from a eutrophic body. Conclusions: Variation in the cellular concentration of cyanobacteria in this reservoir is highly influenced by rain, water mixing and nutrient content.

ABSTRACT

Os aminoácidos tipo micosporinas (MAAs) são compostos, produzidos por algumas espécies de cianobactérias e outros microorganismos, principalmente quando são expostos a radiação ultravioleta (UVR). Estes compostos, que vêm demonstrando funções fotoprotetoras e antioxidantes, têm sido pesquisados para aplicação em protetores solares e em produtos antienvelhecimento. O presente estudo focou na caracterização de cianobactérias e outros organismos quanto à produção de MAAs com potencial aplicação em cosméticos. Neste estudo foram desenvolvidos diversos métodos para identificação (via HPLC-DAD-MS/HRMS), purificação (via HPLC-DAD) e quantificação de MAAs (via LC-MS/MS). Pelo método de identificação de MAAs verificou-se que, das 75 cianobactérias estudadas, 27 cepas (38%) sintetizam MAAs. A cepa Oscilatoria sp. CMMA 1600 produziu a maior diversidade de MAAs. 10 MAAs diferentes foram identificados incluindo um MAA de massa molecular 316 Da. Através de dados espectroscópicos obtidos via HPLC-DAD-HRMS e RMN 1D e 2D confirmou-se que se tratava da micosporina-glicina-alanina. A biossíntese natural deste composto por cianobactérias foi relatada pela primeira vez neste estudo. Quanto à quantificação de MAAs, o protocolo de extração otimizado possibilitou uma excelente recuperação dos compostos de interesse, além de ser bastante simples e não utilizar solventes poluentes. As análises via LC-MS/MS foram realizadas através de experimentos de MRM em modo positivo usando uma coluna de fase reversa. O método validado permitiu determinar e quantificar com precisão os MAAs porphyra-334, shinorina e micosporina-glicina-alanina em corridas de apenas 6 minutos, com limites de deteção inferiores a 0,005 µg.mg -1. Aplicando o método de LC-MS/MS realizaram experimentos de indução de MAAs através de exposição à UVR tendo-se observado um aumento da concentração de MAAs nas cepas que já sintetizam estes compostos e, outras cepas começaram a produzir pelo menos um MAA. As cepas de S. torques-reginae (ITEP-024 e ITEP-026) produziram a maior concentração de MAAs. A cepa ITEP-024 foi ainda exposta a diferentes radiações tendo-se observado que a UVB é que mais influencia a produção de MAAs. Neste estudo foi demonstrado o potencial das cianobactérias como produtores de MAAs que podem ser utilizados como fotoprotores em protetores solares


Mycosporines and mycosporine-like amino acids (MAAs) are UV-absorbing compounds produced by cyanobacteria and other organisms, especially upon exposer to solar ultraviolet radiation (UVR). These compounds are photoprotective and some have additional antioxidant functíons useful to the natural cosmetics market. This study aims to identify MAAs-producing cyanobacteria with potential applicatíons in cosmetics. A HPLC-DAD-MS/HRMS method for the identification of MAAs was developed. Out of the 75 cyanobacteria studied, 27 strains (38%) synthesized MAAs. Oscilatoria sp. CMMA 1600, from homocyte type, produced the greatest diversity of MAAs. 10 different MAAs were identified including a MAA with molecular weight of 316 Da. The chemical structure of mycosporine-glycine-alanine was confirmed by 1D/2D NMR and HRMS analyses. This compound has never been reported from a natural source. In this study, a validated LC-MS/MS quantification method for MAAs is also presented. An easy-to-handle and rapid extraction procedure was developed which uses only water and volatile additives as the extractor solvents. The LC-MS/MS method was performed using multiple reaction monitoring in positive mode with a reverse-phase column. The method enabled the accurate determination and quantification of the MAAs porphyra-334, shinorine and mycosporine-glycine-alanine in a 6 minutes running time, with limits of detection < 0.005 µg.mg-1. MAAs induction experiments were performed through UVR exposure. MAAs are constitutively produced by some cyanobacteria and production was further enhanced following UVirradiance. Other strains start to produce at least one MAA after UV-irradiance. Sphaerospermopsis torques-reginae strain (ITEP-024 and ITEP-026) produced the highest concentration of these photoprotective compounds. S. torques-reginae ITEP 024 strain was further exposed to different radiation compositíons. MAAs were significantly influenced by UVB. In this study, the potential of cyanobacteria as MAA producers, that can be used as photoprotectors in sunscreens, has been demonstrated

ABSTRACT

Este trabajo tuvo como objetivo evaluar el efecto de rotaciones arroceras sobre la población de cianobacterias durante el ciclo de cultivo. El estudio se condujo en Entre Ríos (Argentina). Se evaluaron cuatro rotaciones: arroz-soja (AS); arroz-soja-maíz-soja (ASMS): arroz-arroz (AA) y arroz-pradera (AP). Muestras de suelo y agua se tomaron en cuatro estados del cultivo: implantación, macollaje, panoja embuchada y madurez fisiológica. El número potencial de cianobacterias fue menor en relación a otras áreas arroceras. Los menores recuentos se registraron en el período de implantación y los mayores en el de panoja embuchada. El análisis estadístico no mostró efecto de las rotaciones sobre el número de cianobacterias, aunque AA presentó la mayor abundancia. Se reconocieron un total de 13 géneros de cianobacterias Lyngbya, Oscillatoria,Plectonema, Spirulina, Anabaena, Nostoc, Aulosira, Calothrix, Gloeotri-chia, Aphanocapsa,Croococcus, Mycrocystis y Gloeocapsa. La rotación AP registró la riqueza de géneros.Las rotaciones AS y AA presentaron mayor proporción de cianobacterias filamentosas. Los valores de biodiversidad de Simpson fueron bajos. Se concluyó que las rotaciones arroceras evaluadas tuvieron efecto sobre la proporción de cianobacterias unicelulares a filamentosas, los géneros presentes y la riqueza.


The aim of this study was to evaluate the effect of different rice rotations on cyanobacteria populations during the growing season. The study was conducted in Entre Ríos (Argentina). Four crop rotations were evaluated: rice-soybean (AS); rice-soybean-corn-soybean (ASMS); rice-rice (AA) and rice-pasture (AP). Soil and water samples were collected in 4 stages: implantation, tillering, booting and physiological maturity. The potential number of cyanobacteria population was lowerin relation to other rice sites previously reported. The lowest counts were recorded during implantation whereas the highest counts were obtained during booting. The statistical analysis showed no effect of rotation on the variable number of cyanobacteria, although AA was the most abundant. A total of 13 taxa were identified: Lyngbya, Oscillatoria, Plectonema, Spirulina, Anabaena, Nostoc, Aulosira, Calothrix, Gloeotrichia, Aphanocapsa, Croococcus, Mycrocystis and Gloeocapsa. AP rotation recorded the lowest number of genera. AS and AA had a higher proportion of filamentous cyanobacteria. Simpson's index was low. It was concluded that crop rotations had a differential effect on some aspects of cyanobacteria population such as the proportion of unicellular to filamentous cyanobacteria, genera presence and richness.

Abstract This study aimed to evaluate nutrients concentration and spatial-temporal changes in phytoplankton biovolume during an experimental fish culture in net cages in a lateral arm of Salto Caxias reservoir, Brazil. Two sampling stations were placed in the affected lateral arm and other two in a cageless lateral arm. Neither abiotic variables nor phytoplankton biovolume presented significant differences between the treatments. Only temporal changes were confirmed by the analysis performed. Both lateral arms were classified as oligotrophic, reflecting low influence of the net cages. Phytoplankton growth seems to be limited by nitrogen. Biovolume values were, in general, low and five major functional groups were recognized (E, F, G, K and P). In summer higher biovolume values were observed and representatives of Chlorophyceae and Cyanobacteria belonging to the functional groups F and K, respectively, were the most important. In winter phytoplankton was mainly composed by Bacillariophyceae taxa from P group. G group was also restricted to winter and E group occurred in winter and summer. The variations recorded in phytoplankton structure appear to have been mainly influenced by seasonal changes in temperature, precipitation and nutrients availability. The effects of net cages on the abiotic variables and phytoplankton biovolume appear to have been small, probably due to the small number of net cages employed and the system dilution capacity. However, a permanent monitoring of phytoplankton is recommended, since this environment has a carrying capacity, from which the trophic state may increase.


Resumo Este estudo objetivou avaliar alterações nas variáveis abióticas e no biovolume fitoplanctônico durante o cultivo experimental de peixes em tanques-rede em um braço lateral do reservatório de Salto Caxias, Brasil. Foram selecionadas duas estações de amostragem no braço com tanques-rede e outras duas em um braço sem tanques. As variáveis abióticas e o biovolume fitoplanctônico não apresentaram diferenças significativas entre os locais estudados. Apenas mudanças temporais foram confirmadas pelas análises utilizadas. Os dois braços laterais foram classificados como oligotróficos, refletindo a baixa influência dos tanques-rede. O crescimento do fitoplâncton parece ter sido limitado principalmente por nitrogênio. Os valores de biovolume foram, em geral, baixos e cinco principais grupos funcionais foram observados (E, F, G, K e P). No verão, os maiores valores de biovolume foram observados e representantes de Chlorophyceae e Cyanobacteria dos grupos funcionais F e K, respectivamente, se destacaram. No inverno, o fitoplâncton foi composto principalmente por táxons de Bacillariophyceae do grupo P. O grupo G também foi restrito ao inverno e o grupo E ocorreu no inverno e verão. As variações registradas na estrutura do fitoplâncton parecem ter sido principalmente influenciadas pelas mudanças sazonais de temperatura, precipitação e disponibilidade de nutrientes. Os efeitos dos tanques-rede sobre as variáveis abióticas e biovolume fitoplanctônico parecem ter sido pequenos, provavelmente devido ao pequeno número de tanques utilizados e a capacidade de diluição do sistema. Entretanto, o monitoramento permanente do fitoplâncton é recomendado, uma vez que este ambiente possui uma capacidade de suporte, a partir da qual o estado trófico pode aumentar.

ABSTRACT

The genus Leptolyngbya comprises filamentous cyanobacteria that are important in rice fields. In the rhizosphere, cyanobacteria produce a variety of secondary metabolites such as auxins that are important in agriculture soil performance. To assess this, Leptolyngbya strain MMG-1, was isolated from the rhizosphere of rice plants and described. For this, the morphology of this strain was studied by light microscopy as well as by confocal laser scanning microscopy. Besides, the ability of this strain to synthesize an auxin-like bioactive compound was demonstrated under various culture conditions (different amounts of tryptophan; pH; different alternating light:dark periods; duration of the incubation). The auxin-like compound was extracted from the culture of Leptolyngbya strain MMG-1 and identified as indole-3-acetic acid (IAA) by thin layer chromatography (TLC) as well as by high performance liquid chromatography (HPLC). Our results showed that the strain required the precursor L-tryptophan for the synthesis of IAA. Leptolyngbya strain MMG-1 accumulated IAA intracellularly. The IAA secreted by Leptolyngbya strain MMG-1 was significantly correlated with the initial concentration of L-tryptophan in the medium, as well as with the duration of the incubation. The bioactivity of the secreted IAA was determined by its effect on the rooting pattern of Pisum sativum seedlings. The culture supernatant of Leptolyngbya strain MMG-1 stimulated the seedling lateral rooting, while it decreased root length. Hence, rhizospheric Leptolyngbya produced auxin under different conditions and affected the plants rooting pattern. Rev. Biol. Trop. 62 (3): 1251-1260. Epub 2014 September 01.


El género Leptolyngbya comprende cianobacterias filamentosas que son importantes en los campos de cultivo de arroz. En la rizosfera, las cianobacterias producen una variedad de metabolitos secundarios, tales como auxinas, que son importantes en el rendimiento de la agricultura del suelo. La cepa Leptolyngbya MMG-1, fue aislada de la rizosfera de plantas de arroz y se describe en este trabajo. La morfología de esta cepa se estudió por microscopía de luz, así como por microscopía confocal de barrido láser. Además, se estimó la capacidad de esta cepa para sintetizar el compuesto bioactivo auxina como se demostró en diversas condiciones de cultivo (diferentes cantidades de triptófano; pH; diferente luz alterna: períodos oscuros; duración de la incubación). La auxina se extrajo a partir del cultivo de la cepa Leptolyngbya MMG-1 y se identificó como ácido indol-3-acético (AA) por cromatografía de capa fina (TLC), así como por cromatografía líquida de alta resolución (HPLC). Nuestros resultados mostraron que la cepa requiere el precursor de L-triptófano para la síntesis de IAA. La cepa Leptolyngbya MMG-1 acumula intracelularmente IAA. El IAA secretada por la cepa Leptolyngbya MMG-1 se correlacionó significativamente con la concentración inicial de L-triptófano en el medio, así como con la duración de la incubación. La bioactividad de la IAA secretada se determinó por su efecto sobre el patrón de enraizamiento de plantas de semillero de Pisum sativum. El sobrenadante del cultivo de la cepa Leptolyngbya MMG-1 estimuló el enraizamiento lateral en la plántula, mientras que se redujo la longitud de la raíz. Por lo tanto, la producción de auxina por Leptolyngbya rizosférica afectó el crecimiento de las plantas.

ABSTRACT

In lentic water bodies as reservoirs occur eutrophication processes, originated mainly from human activities (i.e. agriculture, animal exploitation). This influx of nutrients in aquatic ecosystems could promote blooms of potentially toxic cyanobacteria. The purpose of this work is to detect the presence of cyanobacteria strains in water samples, using molecular techniques to help in preventive management of reservoirs dedicated to water purification. We used two molecular techniques to detect genes implied with the synthesis of hepatotoxic microcystins from potentially toxic cyanobacteria strains, and to evaluate the molecular diversity of cyanobacteria in water samples from two high-mountain reservoirs used for purification of drinking water for the metropolitan area of Medellin, Colombia. Between 2010-2011 collections of 12 water samples were taken and DNA extraction together with PCR and DGGE analyses where carried out. We amplified 22 sequences between 250-300bp of the genes mcyA and mcyE, and these sequences were related with several strains and cyanobacteria genera accessions from NCBI-GenBank databases. Moreover, sequence amplifications of the 16S small ribosomal RNA subunit - 16S rRNA- between 400-800bp were also performed in order to use them for the DGGE technique. The amplification products of DGGE were set in polyacrilamide gel with posterior denaturing electrophoresis, and the scanned images of the gel bands were analysed with the software GelCompar II. For Riogrande II and La Fe reservoirs we found 35 and 30 different DGGE bands, respectively, as a measurement of molecular diversity in these artificial ecosystems. Here, we demonstrated the utility of two molecular techniques for the detection of genes associated with toxicity and molecular diversity of cyanobacteria in reservoirs destined for drinking water in urban centers. We recommend strongly following with periodically molecular biology studies in these ecosystems combined with limnological and ecological data, as new tools for management of plants of water purification and for the prevention of potentially public health concerns. En embalses, la eutrofización es consecuencia de procesos naturales y de actividades humanas, lo cual puede facilitar la aparición de afloramientos de cianobacterias potencialmente tóxicas. En este trabajo, se utilizaron dos técnicas moleculares en la detección de genes presentes en cepas potencialmente tóxicas de cianobacterias y evaluaciones de diversidad molecular de cianobacterias en los embalses para potabilización de agua, Riogrande II y La Fe, Colombia. Entre 2010-2011, se tomaron 12 muestras de agua en ambos embalses y se realizaron extracciones de ADN para un análisis de marcadores moleculares mediante PCR y DGGE. Se amplificaron secuencias entre 250-300pb de los genes mcyA y mcyE, implicados en la toxicidad de cepas de cianobacterias. Asimismo, se amplificaron secuencias de la región 16S del ARN ribosomal (422pb), para la técnica DGGE. Se corrieron geles de poliacrilamida en gradientes de desnaturalización, se realizó agrupamiento genético (UPGMA), y se separaron por patrones de bandeo las muestras de cada embalse evaluado. Se demuestra la utilidad de las técnicas moleculares en estudios relacionados con la búsqueda de genes asociados con toxicidad y diversidad molecular de cianobacterias en muestras de agua provenientes de embalses de agua con fines de potabilización para centros urbanos.

ABSTRACT

The knowledge of the biodiversity of Gorgona, a continental island in Colombia, is very limited in the case of algae. We present an updated list of the benthic marine algae of Gorgona Island, associated with different marine environments and type of substrates, such as coral reefs, rocks, and soft bottoms. Field samplings were taken between October 2010 and June 2011. We estimated algal cover, species composition and algal diversity for each reef environment using 0.25 m² quadrats. Ecological and environmental aspects are also included. A total of 43 species of algae for the three types of habitats are documented: 24 Rhodophyta, 12 Clorophyta, six Ochrophyta and one species of Cyanobacteria, and 55% of the algal composition corresponded to red algae. Se presenta el listado de las algas marinas bénticas del Parque Nacional Natural Isla Gorgona, el inventario corresponde a la flora algal asociada a los diferentes ambientes marinos de la isla, a saber: arrecifes coralinos y fondos blandos. El monitoreo de las comunidades de algas se llevó a cabo entre octubre de 2010 y mayo de 2011, se realizó una estimación de abundancia y cobertura para lo cual se establecieron transeptos paralelos a la línea de costa y se evaluó el porcentaje de cobertura en cuadrantes de 0.25m². Se reportó un total de 43 especies para los dos ambientes estudiados (coralino y fondos blandos), 24 Rhodophyta, 12 Clorophyta, 6 Ochrophyta y una Cyanobacteria. El grupo más representativo fue el perteneciente a la clase Rhodophyceae, que correspondió al 55.8% de la flora inventariada. Los resultados de nuevos registros en la isla ratifican su importancia fitogeográfica, por cuanto la composición florística insular contribuye significativamente a la biodiversidad de la costa pacífica colombiana, destacándose la presencia de arrecifes coralinos como los principales ambientes marinos para el desarrollo de la flora algal de la región.

ABSTRACT

Se comparó la eficiencia de sistemas de cultivos discontinuos alimentados versus cultivos discontinuos convencionales, en cuanto a concentración de nitrógeno, adicionando 0,2 mM de urea cada tres días al final de la fase exponencial, durante 21 días. Se realizaron cultivos con un volumen de 1500 mL a 15 y 35 UPS de salinidad, enriquecidos con medio ALGAL 8mM NaNO3, a 238 µmol q m-2 s-1, aireación constante, fotoperiodo 12:12 horas y temperatura de 29 ±3°C. Phormidium sp. posee la capacidad de hidrolizar la urea; mostrando una asimilación de 65±7,07% de la misma, con la mayor producción (p<0,05) de clorofila a, ficocianina y proteínas de 20,26±1,24; 203,47±12,83 y 707,87±28,47 µg mL-1en los cultivos alimentados. La producción de pigmentos vario en el tiempo, independientemente a la salinidad y sistema de cultivo, mientras que la producción de proteínas y carbohidratos totales fue directamente proporcional a la edad del cultivo, con valores máximos de 612,74 ± 5,41 µg mL-1 y 8,96±0,08 mg mL-1 respectivamente a los 31 días. La síntesis de lípidos y EPS fueron influenciadas (p<0,05) por la salinidad, presentando los máximos de lípidos a 15 UPS con 12,22±2,91µg mL-1, y los EPS se incrementaron a 35 UPS con 2,00 ± 0,26 y 2,03 ± 0,15 mg mL-1. Estos resultados determinan que los cultivos de Phormidium sp. alimentados con urea y a salinidades de 15 y 35 UPS, representan una alternativa económica para la producción de clorofila a, ficocianina y proteínas, incrementándose un 31,04; 40,72 y 31,94 % respectivamente en comparación con cultivos no alimentados.


Fed-batch system efficiency versus batch cultures was compared in relation to nitrogen concentration, adding 0,2mM urea at the end of the exponential phase, during 21 days. Cultures were carried out in 1500 mL to 1.5 and 3.5 UPS of salinity, enriched with Algal medium 8mM NaNO3, 238 mol q m-2 s-1, constant aeration, photoperiod 12:12 h. and 29 ±3°C. Phormidium sp. is able to hydrolyze urea; showing a total assimilation of 65±7.07%, with the highest (p< 0.05) chlorophyll a, phycocyanin and protein production of 20.26 ± 1.24, 203.47 ± 12.83 and 707.87 ± 28.47 µg mL-1 in the fed-batch cultures. On the other hand, pigment production varies in time, regardless salinity and culture system. Proteins and total carbohydrate production were directly proportional to the age of cultures, with maximum values of 612.74 ± 5.41 µg mL-1 and 8.96 ± 0.08 mg mL-1, respectively. Lipid and EPS were influenced (p< 0.05) by salinity, showing maximum of lipids at 15 UPS with 12.22±2.91 µg mL-1, and EPS at 15 and 35 UPS with 2.00 ± 0.26 and 2.03 ± 0.15 mg mL-1. These results determine that Phormidium sp. cultures fed with urea, to salinities of 15-35 UPS, represent an economic alternative for chlorophyll a, phycocyanin and protein production, with an increase of 31.04, 40.72 and 31.94% respectively in comparison with non-fed cultures.

ABSTRACT

The tropical karstic lakes on the Mexican Caribbean Sea coast are numerous. However, there is an enormous gap of knowledge about their limnological conditions and micro-algae communities. In the present study, surface water samples were collected monthly from November 2007 to September 2008 to provide taxonomical composition and biovolume of planktonic cyanobacteria of the lake Lagartos from State of Quintana Roo, Mexico. Water temperature, pH, conductivity, salinity, soluble reactive phosphorus (SRP), dissolved inorganic nitrogen (DIN), and soluble reactive silica (SRSi) levels were also analyzed. A total of 22 species were identified. Chroococcales and Oscillatoriales dominated the phytoplankton assemblages during the study period. Chroococcus pulcherrimus, Coelosphaerium confertum, Cyanodyction iac, Phormidium pachydermaticum and Planktolyngbya contorta were recorded for the first time in Mexico. A surplus of DIN (mean value of 42.7µM) and low concentrations of SRP (mean value of 1.0µM) promoted the enhanced growth and bloom formation of cyanobacteria. The mean biovolume was 3.22X10(8)µm³/mL, and two biovolume peaks were observed; the first was dominated by Microcystis panniformis in November 2007 (7.40X10(8)µm³/mL), and the second was dominated by Oscillatoria princeps in April 2008 (6.55X10(8)µm³/mL). Water quality data, nitrates enrichment, and trophic state based on biovolume, indicated that Lagartos is a hyposaline, secondarily phosphorus-limited, and eutrophic lake, where the cyanobacteria flora was composed mainly by non-heterocystous groups.


Los lagos cársticos tropicales en la costa del Caribe mexicano son numerosos. Sin embargo, existe un enorme desconocimiento acerca de sus condiciones limnológicas y de las comunidades de microalgas que se desarrollan en ellos. El objetivo del presente estudio fue estudiar las condiciones limnológicas en las que crecen las poblaciones de cianobacterias planctónicas del lago Lagartos, Quintana Roo, México. Las recolectas se realizaron de forma mensual entre noviembre 2007 y septiembre 2008. Las especies fueron identificadas y su biovolumen determinado. Se midieron in situ la temperatura del agua, pH, conductividad y salinidad. También, se analizaron las concentraciones de fósforo reactivo soluble (SRP), nitrógeno inorgánico disuelto (DIN) y sílice reactivo soluble (SRSi). Se identificaron 22 especies de cianobacterias. Chroococcus pulcherrimus, Coelosphaerium confertum, Cyanodyction iac, Phormidium pachydermaticum y Planktolyngbya contorta fueron nuevos registros para México. Un exceso de DIN (valor promedio de 42.7µM) y bajas concentraciones de PRS (valor promedio de 1.0µM) promovieron la proliferación de cianobacterias. El biovolumen presentó dos picos: el primero en noviembre 2007, dominado por Microcystis panniformis (7.40X10(8)µm³/mL) y el segundo en abril 2008, representado por Oscillatoria princeps (6.55X10(8)µm³/mL). Los datos de calidad del agua, el enriquecimiento por nitratos y el estado trófico basado en el biovolumen, indican que Lagartos es un lago hiposalino, eutrófico, con limitación secundaria por fósforo, donde los crecimientos masivos de cianobacterias sin heterocitos fueron recurrentes.

Microbial biomass and activity in soils are frequently studied in tropical dry forests, but scarce information is available about the relationships between functional bacterial groups and soil fertility, where relief interacts with rainfall seasonality. The culturable-bacterial groups and nutrients were studied during two consecutive years in soils from two topographic areas of different relief (hilltop vs hillslope) in a tropical dry forest from Chamela Jalisco, Mexico. We expected that seasonal and spatial variation in soil resources availability affects the abundance of functional culturable-bacterial groups. To evaluate this, fifteen soil cores (1kg), 0-5cm depth, were taken in the dry, early rainy and rainy seasons, from each of the ten replicate plots in hilltop and hillslope areas located in three microbasins. We found that hilltop soils were more organic and had higher concentration of labile C and total nutrient forms than hillslope soils, for which these soils had higher counts of colony-forming units (CFU) of total heterotrophic and P solubilizing bacteria. In both hilltop and hillslope soils, C and nutrient concentrations, as well as the counts of CFU of heterotrophic and P solubilizing bacteria generally decreased from the dry to the rainy season during the two study years. In contrast, the counts of CFU nitrifying and cellulolytic bacteria were higher at the hillslope than at the hilltop soils. The seasonal pattern of both groups was opposite to that of heterotrophic bacteria, presumably associated with a decrease in soil labile C and organic matter quality. In conclusion, our study suggests that available C appears to be the main factor that controls the structure of soil bacterial groups and soil fertility, where relief, rainfall seasonality and intraand inter-annual variations are critical factors that interactively modify bacterial dynamics related to soil C availability in the tropical dry forest.


Los bosques tropicales secos tienen una amplia heterogeneidad ambiental y por lo tanto se podría esperar una variación temporal y espacial en la abundancia y tipo de grupos bacterianos activos. Se evaluó, durante dos años consecutivos, el efecto de la estacionalidad de la lluvia sobre algunos grupos bacterianos cultivables y nutrimentos del suelo en dos posiciones topográficas (crestas vs laderas) en el bosque tropical seco, Chamela Jalisco, México. La hipótesis fue que la variación temporal y espacial de la disponibilidad de recursos afectaría la abundancia de los grupos bacterianos funcionales (heterótrofas totales, celulolíticas, solubilizadores de fosfatos y nitrificantes) del suelo. En las crestas, el suelo fue más orgánico y con mayor concentración de nutrimentos totales y C orgánico lábil que en la ladera, registrando más bacterias heterótrofas y solubilizadoras de fosfato. En ambas posiciones topográficas la concentración de C y nutrimentos, así como el número de bacterias heterótrofas y solubilizadoras de P, decrecieron de la estación seca a la lluviosa en ambos años. En contraste, las bacterias nitrificantes fueron mayores en el suelo de la ladera, y al igual que las celulolíticas, tuvieron un patrón estacional opuesto a las bacterias heterótrofas, asociado a la disminución del C lábil y de la calidad de la materia orgánica. En conclusión, se sugiere que el relieve, la estacionalidad y la variación inter-anual de la lluvia regulan interactivamente la relación entre el C disponible y la estructura de las comunidades bacterianas del suelo en el bosque tropical seco de Chamela.

ABSTRACT

Aerophytic cyanobacteria are commonly found growing on rocks, tree trunks and soil, but the diversity of these organisms is still poorly known. This complex group is very problematic considering the taxonomic arrangement and species circumscription, especially when taking into account tropical populations. In this work, 20 samples of aerophytic cyanobacteria from 15 distinct sampling sites were collected along a tropical rainforest area at the São Paulo State (Brazil). Samples were dried at room temperature after the collection, and once in the laboratory, they were rehydrated and analyzed. The taxonomic study resulted in the record and description of nine species of true-branched cyanobacteria pertaining to the genera Spelaeopogon (one specie), Hapalosiphon (two species) and Stigonema (six species). The similarity of the flora found when compared to surveys conducted in other geographical regions was relatively low. These differences could be addressed to ecological conditions of the habitats, to the extension of the area surveyed or even to taxonomic misinterpretations. The molecular assessment of 16S rDNA on the basis of field material was successful for two morphospecies, Hapalosiphon sp. and Stigonema ocellatum; however, their relationships with other populations and species revealed to be uncertain. The results of the floristic survey and of the molecular approach evidenced the fragile delimitation of some genera and species in the true branched group of cyanobacteria.


Las bacterias aerofíticas crecen comúnmente en las rocas, troncos de árboles y el suelo, pero la diversidad de estos organismos todavía es poco conocida. Este complejo grupo es muy problématico considerando la disposición taxónomica y la circunscripción de las especies, especialmente cuando se toman en cuenta las poblaciones tropicales. En esta investigación, 20 muestras de cianobacterias aerofíticas de 15 distintos sitios de muestro fueron recolectadas a lo largo de un área de selva tropical en el estado de São Paulo (Brasil). Las muestras fueron secadas a temperatura ambiente después de su recolecta, y una vez en el laboratorio, se hidrataron y analizaron. El estudio taxónomico resultó en un registro y descripción de nueve especies de cianobacterias con ramificaciones verdaderas pertenecientes al género Spelaeopogon (una especie), Hapalosiphon (dos especies) y Stigonema (seis especies). La similutud de la flora encontrada en comparación con las encuestas realizadas en otras regiones geográficas fue relativamente baja. Estas diferencias podrían deberse a las condiciones ecológicas de los hábitats, a la extensión de la zona de estudio o incluso a malas interpretaciones taxonómicas. La evaluación molecular del 16S rDNA del material de campo fue un éxito para dos morfoespecies, Hapalosiphon sp. y Stigonema ocellatum, sin embargo, sus relaciones con otras poblaciones y especies revelan que son inciertas. Los resultados del estudio florístico y de la aproximación molecular evidencian la débil delimitación de algunos géneros y especies en el grupo de cianobacterias con ramificaciones verdaderas.

This study aimed to analyse the composition and ecological attributes of the phytoplankton assemblages in four lateral lagoons and in the main channel of Rosana Reservoir (Paranapanema River, SE Brazil). Fieldwork was carried out in September and November/2004 and January, March, May and August/2005. A total of 283 taxa was identified. Zygnemaphyta was the most specious group, followed by Chlorophyta and Bacillariophyta. Higher richness, abundance and biomass were observed in the lagoons when compared with the river-reservoir sampling point, especially during the rainy period. Cryptophyceae and Bacillariophyceae dominated numerically. Cryptomonas brasiliensis Castro, Bicudo and Bicudo was the main species of the phytoplankton in terms of abundance and frequency of occurrence. The dynamics of the most important taxa are discussed and the results showed that the phytoplankton assemblages are mainly influenced by meteorological factors and nutrient availability (the main driving forces). Correlation analyses indicated that the assemblage abundance was limited by nutrient (nitrogen and phosphorus). The phytoplankton abundance influenced positively the zooplankton abundance, what indicates the prevalence of bottom-up control routes in the lateral lagoons system. The results validate the hypotheses that lateral lagoons have a prominent ecological role on the phytoplankton diversity, as already previously demonstrated for fish and zooplankton. Therefore, the incorporation of the lateral lagoons in environmental programmes should be a target strategy for the conservation of the regional aquatic biota, minimising the negative impact of the dam.


O objetivo deste estudo foi analisar a composição e os atributos ecológicos das assembleias fitoplanctônicas de quatro lagoas marginais e canal principal do Reservatório de Rosana (SE, Brasil). Os trabalhos de campo foram realizados em setembro e novembro de 2004, e janeiro, março, maio e agosto de 2005. O número total de táxons identificados foi de 283. Zygnemaphyta foi o grupo com maior número de espécies, seguido por Chlorophyta e Bacillariophyta. Maior riqueza, abundância e biomassa foram observadas nas lagoas, quando comparadas ao canal do rio-reservatório, especialmente durante o período chuvoso. Cryptophyceae e Bacillariophyceae dominaram numericamente as assembleias. Cryptomonas brasiliensis Castro, Bicudo e Bicudo foi a principal espécie do fitoplâncton, em termos de abundância e frequência de ocorrência. Os resultados mostraram que as assembleias fitoplanctônicas são influenciadas principalmente pelos fatores meteorológicos e nutrientes (funções de força). Análises de correlação mostraram que a abundância do fitoplâncton foi limitada por nutrientes (nitrogênio e fósforo). Por sua vez, a abundância fitoplanctônica influenciou positivamente a abundância do zooplâncton, o que indica a predominância de mecanismos de controle ascendente no sistema de lagoas laterais. Os resultados validaram a hipótese, assim como já havia sido demonstrado para peixes e zooplâncton, de que as lagoas laterais têm um proeminente papel na diversidade do fitoplâncton. A incorporação das lagoas laterais em programas ambientais seria uma boa estratégia para a conservação da biota aquática regional, minimizando o impacto negativo do represamento.

Environmental conditions favor the predominance of dense populations of cyanobacteria in reservoirs in northeastern Brazil. The aim of this study was to understand cyanobacterial population dynamics in the rainy and dry seasons at two depths in the Arcoverde reservoir. Microalgae and cyanobacteria samples were collected during 24 hours with intervals of 4 hours (nycthemeral) at sub-surface and 10 m using a van Dorn bottle and a determined biomass. Physical and chemical variables were obtained and the data were analyzed using the principal component analysis (PCA). No nycthemeral variations in the taxonomic composition or distribution of the populations of cyanobacteria were found between the different times of day in either the rainy or dry season. In both seasons, the greatest biomass of the phytoplankton community was made up of cyanobacteria at two depths and all times of the day. Cylindrospermopsis raciborskii (Woloszynska) Seenayya et Subba Raju was dominant at all times of the day on both the surface and at the bottom. In the rainy season, the differences in cyanobacterial biomass between the surface and bottom were less significant than in the dry season. The differences in cyanobacterial biomass between surface and bottom were less pronounced than those found in the dry season. We concluded that a) physical variables better explain the alterations of species in the phytoplankton community in an environment dominated by cyanobacteria throughout the year; b) seasonal climatic factors associated to periods of stratification and de-stratification are important for alterations in the community and variations in biomass and, c) the turbidity caused by rainfall favored the emergence and establishment of other cyanobacteria, especially Planktothrix agardhii (Gomont) Anagnostidis & Komárek.


As condições ambientais favorecem a predominância de populações densas de cianobactérias no nordeste do Brasil. O objetivo deste estudo foi entender a dinâmica das populações de cianobactérias nas estações seca e chuvosa, em duas profundidades, no reservatório de Arcoverde. Amostras de microalgas e cianobactérias foram coletadas durante 24 horas em intervalos de quatro horas (nictemerais) na subsuperfície e em 10 m, com uma garrafa de van Dorn, para a determinação da biomassa. Variáveis físicas e químicas foram obtidas, e os dados investigados por meio da Análise de Componentes Principais (ACP). Não foram encontradas variações nictemerais na composição taxonômica ou na distribuição das populações de cianobactérias entre os diferentes horários do dia, bem como entre as estações seca e chuvosa. Em ambas as estações, horários do dia e profundidades amostrais, a maior biomassa da comunidade fitoplanctônica foi constituída por cianobactérias. Cylindrospermopsis raciborskii (Woloszynska) Seenayya et Subba Raju foi dominante em todos os horários do dia, tanto na superfície como no fundo. Na estação chuvosa, as diferenças de biomassa de cianobactérias entre superfície e fundo foram menos pronunciadas do que na estação seca. Concluímos que: a) variáveis físicas explicam melhor as alterações das espécies na comunidade fitoplanctônica em um ambiente dominado por cianobactérias durante todo o ano; b) fatores climáticos sazonais associados a períodos de estratificação e desestratificação são importantes para as alterações na comunidade e as variações na biomassa, e c) a turbidez causada pela chuva favoreceu o aparecimento e o estabelecimento de outras cianobactérias, especialmente Planktothrix agardhii (Gomont) Anagnostidis & Komárek.

This study investigated the dynamics of cyanobacteria in two deep, eutrophic reservoirs in a semi-arid region of Brazil during periods of stratification and destratification. Four collections were carried out at each reservoir at two depths at three-month intervals. The following abiotic variables were analyzed: water temperature, dissolved oxygen, pH, turbidity, water transparency, total phosphorus, total dissolved phosphorus, orthophosphate and total nitrogen. Phytoplankton density was quantified for the determination of the biomass of cyanobacteria. The data were analyzed using CCA. Higher mean phytoplankton biomass values (29.8 mm³.L-1) occurred in the period of thermal stratification. A greater similarity in the phytoplankton communities also occurred in this period and was related to the development of cyanobacteria, mainly Cylindrospermopsis raciborskii (>3.9 mm³.L-1). During the period of thermal destratification, this species co-dominated the environment with Planktothrix agardhii, Geitlerinema amphibium, Microcystis aeruginosa and Merismopedia tenuissima, as well as with diatoms and phytoflagellates. Environmental instability and competition among algae hindered the establishment of blooms more during the mixture period than during the stratification period. Thermal changes in the water column caused by climatologic events altered other physiochemical conditions of the water, leading to changes in the composition and biomass of the cyanobacterial community in tropical reservoirs.


Este estudo investigou a dinâmica das cianobactérias em dois reservatórios eutróficos e profundos localizados na região semi-árida do Brasil durante períodos de estratificação e desestratificação térmica. Quatro coletas foram feitas em cada reservatório em duas profundidades em intervalo de três meses. As variáveis abióticas analisadas foram: temperatura da água, oxigênio dissolvido, pH, turbidez, transparência da água, fósforo total, fósforo total dissolvido, ortofosfato e nitrogênio total. A densidade do fitoplâncton foi quantificada para a determinação da biomassa de cianobactérias. Os dados foram analisados usando ACC. Elevados valores médios de biomassa fitoplanctônica (29,8 mm³.L-1) ocorreram no período de estratificação. A maior similaridade das comunidades fitoplanctônicas também ocorreu neste período e foi relacionada ao desenvolvimento de cianobactérias, principalmente Cylindrospermopsis raciborskii (>3,9 mm³ .L-1). Durante o período de desestratificação térmica, esta espécie co-dominou o ambiente com Planktothrix agardhii, Geitlerinema amphibium, Microcystis aeruginosa e Merismopedia tenuissima, bem como com diatomáceas e fitoflagelados. Instabilidade ambiental e competição entre as algas dificultaram o estabelecimento de blooms durante o período de mistura quando comparado com o período de estratificação. Modificações na condição térmica da coluna d'água, causadas por eventos climatológicos, alteraram as demais condições fisicoquímicas da água e ocasionaram mudanças na composição e biomassa da comunidade de cianobactérias em reservatórios tropicais.

This study analysed vertical and temporal variations of cyanobacteria in a potable water supply in northeastern Brazil. Samples were collected from four reservoir depths in the four months; September and December 2007; and March and June 2008. The water samples for the determination of nutrients and cyanobacteria were collected using a horizontal van Dorn bottle. The samples were preserved in 4 percent formaldehyde for taxonomic analysis using an optical microscope, and water aliquots were preserved in acetic Lugol solution for determination of density using an inverted microscope. High water temperatures, alkaline pH, low transparency, high phosphorous content and limited nitrogen content were found throughout the study. Dissolved oxygen stratification occurred throughout the study period whereas temperature stratification occurred in all sampling months, with the exception of June. No significant vertical differences were recorded for turbidity or total and dissolved forms of nutrients. There were high levels of biomass arising from Planktothrix agardhii, Cylindrospermopsis raciborskii, Geitlerinema amphibium and Pseudanabaena catenata. The study demonstrates that, in a tropical eutrophic environment with high temperatures throughout the water column, perennial multi-species cyanobacterial blooms, formed by species capable of regulating their position in the water column (those that have gas vesicles for buoyancy), are dominant in the photic and aphotic strata.


O presente estudo analisou as variações vertical e temporal de cianobactérias em um reservatório de abastecimento de água no nordeste do Brasil. As amostras foram coletadas em quatro profundidades no reservatório, durante quatro meses (setembro e dezembro de 2007, março e junho de 2008). As amostras de água para a determinação de nutrientes e de cianobactérias foram coletadas por meio de garrafa horizontal de Van Dorn. As amostras foram preservadas em formol 4 por cento para posterior análise taxonômica utilizando um microscópio óptico, e alíquotas de água foram preservadas em solução de Lugol acético para determinação da densidade através de um microscópio invertido. Altas temperaturas da água, pH alcalino, baixa transparência, alto teor de fósforo e limitação de nitrogênio foram encontrados ao longo do estudo. Durante todo o estudo, foi verificada estratificação do oxigênio dissolvido enquanto que estratificação térmica só não foi observada em junho. Diferenças verticais não foram registradas para a turbidez e formas totais e dissolvidas de nutrientes. Ocorreram altas biomassas de Planktothrix agardhii, Cylindrospermopsis raciborskii, Geitlerinema amphibium e Pseudanabaena catenata. O presente estudo demonstra que, em um ambiente eutrófico tropical com temperaturas elevadas em toda a coluna d'água, florações perenes multiespécies de cianobactérias formadas por espécies capazes de regular a sua posição na coluna de água (aqueles que possuem vacúolos de gás para flutuação) são dominantes nas camadas fótica e afótica.

ABSTRACT

The physicochemical variables and cyanobacteria of Mid-Cross River, Nigeria, were studied in six stations between March 2005 and August 2006 to determine the relationship between water quality and cyanobacteria abundance. Canonical Correspondence Analysis (CCA) showed that biological oxygen demand (BOD), dissolved oxygen, pH, water velocity, width and depth were important environmental factors that influenced cyanobacteria abundance. Trace metals, phosphate and nitrate increased significantly from values of previous studies indicating increased eutrophication of the river but were weakly correlated with cyanobacteria abundance and could be scarcely regarded as regulating factors. A higher cyanobacteria abundance was recorded during the wet season in most of the sampled stations. The dominant cyanobacteria included Microcystis aeruginosa, Aphanizomenon flos-aquae, Oscillatoria limnetica and Anabaena spiroides. The toxins produced by these species could degrade water quality. The factors favouring cyanobacteria abundance were identified as increased pH, width and depth. Increase in cyanobacteria abundance was associated with reduction in dissolved oxygen and increase in BOD values. Rev. Biol. Trop. 57 (1-2): 33-43. Epub 2009 June 30.


Las variables físico-químicas y la abundancia de cianobacterias del río nigeriano Mid-Cross fueron estudiadas en seis estaciones entre marzo del 2005 y agosto del 2006. El Análisis de Correspondencia Canónica (CCA) demostró que la demanda biológica de oxígeno (DBO), oxígeno disuelto, pH, velocidad de agua, anchura y profundidad son factores ambientales importantes que influyen en la abundancia de cianobacterias. Los mayores valores de trazas de metales, fosfatos y nitratos, en comparación con estudios previos, indican mayor eutrofización, pero tienen poca correlación con la abundancia de las cianobacterias. La mayor abundancia de cianobacterias se registró durante el periodo más húmedo de la estación seca en la mayoría de las estaciones de muestreo. Las cianobacterias dominantes incluyen Microcystis aeruginosa, Aphanizomenon flos-Aquae, Oscillatoria limnetica y Anabaena spiroide. Las toxinas producidas por estas especies podrían degradar la calidad del agua. Los factores que favorecen la abundancia de cianobacterias fueron identificados como el aumento de pH, ancho y profundidad. El aumento de la abundancia se asoció con una reducción de oxígeno disuelto y un aumento de los valores de DBO.

Periodical changes in physico-chemical properties and cyanophycean bloom in a pond ofAgra city during the year 2005 and 2006 were studied. It was recorded that the pond water was polluted throughout the study period and this was closely associated with Cyanobacterial bloom. During summer (March to June), the pond water was highly polluted, as it was rich in various nutrients and phosphates in particular. The pH of pond water was high with low DO and this was associated with the highest peak of water bloom. On the other hand, during winter (November to January), the bloom was at its lowest. The quantity of various nutrients in the pond water was low and the pH was also low but DO was high. Bloom consisted of eighteen members of Cyanophyceae of which Oscillatoria limosa, O. formosa, O. splendida, Anabeana iyengarii, Microcystis arruginosa and Nostoc muscorum were observed throughout the year. On the other hand, Oscillatoria agardhi, and O. tenuis were found only in winters, while O. annae in rainy season only. Similarly, Oscillatoria brevis, Anabaena flosaquae, Aphanizomenon sp. and Lyngybea birgei were found in abundance in summer.

ABSTRACT

Field survey of algae and cyanobacteria from terrestrial and freshwater habitats in the vicinity of arctic Ny-Alesund, Svalbard (790N) (high Arctic sea area) was performed in June 2006. Species diversity and abundance were evaluated by using epifluorescence microscopy and culturing methods. In total, 29 taxa in 25 genera were identified, of which Leptolyngbya spp., Trichormus sp. and Chlamydomonas nivalis were abundantly present in almost every sample. In several locations, blooms were formed by species C. nivalis, Scotiellopsis sp., Klebsormidium flaccidum, Zygnema sp., Meridion circulare, Tabellaria fenestrata and Fragilaria sp. Eleven new species from this locality are described.

ABSTRACT

En Chile se ha detectado la presencia de algunos géneros de cianobacterias que pueden producir potentes hepatotoxinas y neurotoxinas, las que pueden ser letales para humanos y animales. En el presente trabajo se determinó la presencia de dos géneros de cianobacterias no tóxicos: Chroococcus y Spirulina; y cuatro géneros de cianobacterias productores de toxina, Anabaena, Anabaenopsis, Microcystis y Oscillatoria en tres diferentes cuerpos de agua de la V Región: Lago Peñuelas (Valparaíso), Tranque Recreo (Villa Alemana) y Embalse Los Aromos (Limache). Además se detectó la presencia de hepatotoxinas por MALDI-TOF MS encontrándose microcistina-RR, -LA, -YR y nodularina en Embalse Los Aromos, microcistina-LA en Tranque Recreo y microcistina-RR y LA en Lago Peñuelas. Adicionalmente en algunas de las muestras se detectó la presencia de péptidos no tóxicos, que presentan actividad biológica tales como aeruginosinas, cianopeptolinas y microgininas. Como estos cuerpos de agua dulce son utilizados para abastecimiento público y para la recreación, es importante diseñar planes de tratamiento y monitoreo para detectar y evitar los riesgos a la salud humana y animal provocado por estos microorganismos.


In Chile the presence of some genera of cyanobacteria that may cause potent hepatoxins and neurotoxis has been detected, which may become lethal for man and animal. In this paper the occurrence of two non toxic genera of cyanobacteria: Chroococcus and Spirulina was established along with four genera of toxin-producing cyanobacteria, Anabaena, Anabaenopsis, Microcystis and Oscillatoria in three different masses of water from the V Region: Lago Peñuelas (Valparaíso), Tranque Recreo (Villa Alemana) and Embalse Los Aromos (Limache). Likewise the presence of hepatoxins by MALDI-TOF MS was detected which resulted in the occurrence of microcystina-RR, _LA , -YR and nodularina in Embalse Los Aromos, microcystina –LA in Tranque Recreo and microcystina -RR and LA in Lago Peñuelas. Moreover, the presence of non toxic peptides developing biological activities such as aeruginosinas, cianopeptolins and microgininines was detected. Considering that these freshwater bodies are intended for public supply and recreational purposes, it is of utmost importance to design treatment and supervising plans in order to detect and prevent risks for human and animal health caused by these microorganisms.
